Re-derivation to the clean units at UCPH

Re-derivation of your line to clean units
As soon as the mice have arrived you can initiate the re-derivation from the quarantine to the clean units. At this point the Core Facility will take over the process and we will keep you informed on the progress of your derivation.
As a standard method, we will re-derive mouse lines by isolating sperm from one male and using it for in vitro fertilization of C57Bl/6N oocytes. Oocytes can also be derived from other mouse strains, if requested. Using this method a large number of embryos can be generated, which will then be transferred to recipient mice at the SPF units. Recipient mice will be housed in isolators and after weaning of the pups, the recipient mothers will be sent for FELASA check and when proven clean the pups are released to the user.
If mice should be housed only at the Biocenter, 16.4 or 10.3, no FELASA check is required and the process will be faster and less expensive.
Combination of re-derivation with sperm cryopreservation
Sperm freezing is an inexpensive and reliable method to archive mouse strains (link to spermfreezing). If you are interested in this archiving - it is most economical to combine it with the re-derivation, since sperm isolation, IVF and embryo transfer are part of both procedures. The sperm freezing will provide you with a safe back up of your line.
